چکیده

Sucralose is a commonly employed artificial sweetener that appears to destabilize protein native structures. This is in direct contrast to the bio-preservative nature of its natural counterpart, sucrose, which enhances the stability of biomolecules against environmental stress. We have further explored the molecular interactions of sucralose as compared to sucrose to illuminate the origin of the differences in their bio-preservative efficacy. We show that the mode of interactions of sucralose and sucrose in bulk solution differ subtly using hydration dynamics measurement and computational simulation. Sucralose does not appear to disturb the native state of proteins for moderate concentrations (<0.2 M) at room temperature. However, as the concentration increases, or in the thermally stressed state, sucralose appears to differ in its interactions with protein leading to the reduction of native state stability. This difference in interaction appears weak. We explored

Sucralose Destabilization of Protein Structure.

Sucralose is a commonly employed artificial sweetener that behaves very differently than its natural disaccharide counterpart, sucrose, in terms of its interaction with biomolecules. The presence of sucralose in solution is found to destabilize the native structure of two model protein systems: the globular protein bovine serum albumin and an enzyme staphylococcal nuclease. The melting temperat...

Cob(I)alamin reacts with sucralose to afford an alkylcobalamin: relevance to in vivo cobalamin and sucralose interaction.

Vitamin B(12), viz., cyano- or hydroxo-cobalamin, can be chemically or enzymatically converted into the derivatives methyl- and adenosyl-cobalamin, which are complex organometallic cofactors associated with several cobalamin-dependent enzymes. The reduced form of vitamin B(12), cob(I)alamin {Cbl(I)}, obtained by reduction of hydroxocobalamin (OH-Cbl) with e.g. sodium borohydride, is one of the ...

Optical and Nano structural properties of Hematite (α-Fe2O3) nanorods in interaction with Bovine Serum Albumin (BSA) Protein Solution

Hematite (α-Fe2O3) nanorods were synthesized by hydrothermal method using Cetyltrimethylammonium bromide (CTAB) as a surfactant agent. To study optical, nanostructural properties, and to control the morphology and shape of nanorods, 0.025 mol L-1, 0.05 mol L-1 and 0.1 mol L-1 concentration of CTAB were used. Moreover, the effect of interaction between bovine serum albumin (BSA) A9418-5G protein...

Sucralose activates an ERK1/2–ribosomal protein S6 signaling axis

The sweetener sucralose can signal through its GPCR receptor to induce insulin secretion from pancreatic β cells, but the downstream signaling pathways involved are not well-understood. Here we measure responses to sucralose, glucagon-like peptide 1, and amino acids in MIN6 β cells. Our data suggest a signaling axis, whereby sucralose induces calcium and cAMP, activation of ERK1/2, and site-spe...

Gut Microbiome Response to Sucralose and Its Potential Role in Inducing Liver Inflammation in Mice

Sucralose is the most widely used artificial sweetener, and its health effects have been highly debated over the years. In particular, previous studies have shown that sucralose consumption can alter the gut microbiota. The gut microbiome plays a key role in processes related to host health, such as food digestion and fermentation, immune cell development, and enteric nervous system regulation....
